Can the effect of one Demise destroy the other?

Of course. There's nothing in Demise, King of Armageddon's text that makes him immune to any kind of monster destruction.

Demise is protected from his own effect. So he's not destroyed when you activate it. However, another Demise, exiled force, a fissure, or whatever destroy monster card can take him out. He's not protected from them.

Also, if you have two on the field, activating the effect of one will destroy the other.
