Currently building an annoying deck based around Ameba and Griggle.

The tactics
I use Shien's Spy to switch control of my face up Ameba and do 2,000 damage. At the end phase Ameba comes back to me.

The question

If on my next turn I summon Tsyukomi and flip Ameba face down, then flip him face up and activate a second Shien's Spy to shift control again, will I do a second 2,000 damage?

My thoughts

I think it works as Ameba says "once only, as long as this card remains face up." The flip down should reset the effect.

Testing results.

Have play tested against a solid beatdown and I drew three monsters 17 magic and traps (mainly magics), almost wet my self laughing as I creature swapped, snatched and brain controlled my way to victory. Opponent could not duel to to hysterical laughter.
 
I used to have a deck like that, implementing Shien's Spy and Creature Swap. Both cards come in handy at certain times. Kind of the only annoying part of that strategy.

But, yes, your idea works. I used the same thing. :D
 
Personally I've been using Mystic Box and Creature Swap....and my deck uses 2 copies of Penguin Soldier and 2 copies of Compulsory Evactuation Device to reclaim Ameba....as well as a copy of Pot Of Avarice. I also use 1 copy of Lava Golem and side deck another. It's actually a reasonably competitive deck....not a tournament winner mind you but it's not unsual to go 3-1 with it.
 
I run the box in my Giant Kozaky deck. Granted I need the face down aspect of the Box over Shien's Spy, but doing the "double whammy" as well works wonders, take out one of their monsters AND give them your little friend to hurt them.

Have you thought about tossing in a Giant Germ set into the deck? You can do a similar tactic of swapping and then attacking, which will still do damage to your opponent and net you a some more swap/stall material since the effect doesn't activate until it hits the graveyard.
 
Yes, when he gets flipped face down his effect gets reset. So your opponent could Tsuku him and flip him back up and then swap him back to you.

I use Creature Swap, Mystic Box, and Shien's spy to give my opponent Ameba. I use Penguin Soldier as well as salvage to give me another shot at using him again. I also use Tsuku to reset him or MoF. My favorite combo on an open field is to summon Ameba, Shien spy him Then special summon Cyber Dragon and attack. Here is my deck
